    1、Sperm DNA fragmentation test

    Meaning of the test

    Technology Highlights

    Damaged sperm chromatin formed single-stranded DNA after acidification, and the single-stranded DNA combined with the dye acridine orange showed orange or red fluorescence by 488 nm laser excitation; whereas normal sperm chromatin retained the intact double-stranded structure of DNA, and combined with the dye acridine orange showed green fluorescence by 488 nm laser excitation. An increase in the percentage of red light after acidification of semen samples indicates an increase in chromatin structure abnormalities.

    Meaning of the test

    ● Assessing sperm motility and fertilization potential: Sperm mitochondrial function directly impacts energy supply for sperm movement; detecting its activity (e.g., membrane potential) evaluates sperm vitality and fertilization capability.

    ● Diagnosing causes of male infertility: Mitochondrial dysfunction is a key factor in asthenozoospermia or azoospermia; testing helps identify metabolic origins of infertility.

    ● Optimizing assisted reproductive techniques (ART): Selecting sperm with intact mitochondrial function for ICSI may enhance embryonic development success rates.

     

    Technology Highlights

    JC-1 is a carbocyanine-based fluorescent dye used to detect the mitochondrial membrane potential ΔΨm, and exists in two different forms, polymers (J-aggregates) and monomers (monomers), in the cell. When the mitochondrial membrane potential is high, JC-1 accumulates in the mitochondrial matrix and forms polymers, which can produce orange-red fluorescence. When the mitochondrial membrane potential is low, JC-1 can not be aggregated in the mitochondrial matrix, and then JC-1 is a monomer, which can produce green fluorescence.

     

    Features:

    Adopt international JC-1 probe.

    Report the percentage of sperm with high mitochondrial membrane potential with significant clinical application.

    Required equipment: flow cytometer (blue excitation).

    Technology Highlights

    The acrosome reaction is a key process in the fertilisation of sperm and egg, including the fusion of plasma membrane and acrosomal membrane, secretion of enzymes, etc. If the acrosome reaction is insufficient or abnormal, it will lead to male infertility. Sperm without acrosome reaction may have a complete acrosome, or it may have a missing acrosome that does not fluoresce. And sperm with acrosome reaction exposed CD46 antigen on the acrosome membrane, which specifically binds to FITC conjugated CD46 antibody and emits green fluorescence.
